After Julian Speroni failed to hold James Ward-Prowse's shot, the Senegalese chipped over the Palace keeper to score the winner for Southampton.
The visitors had looked most likely to score with Wilfried Zaha striking a post in the second half, but a first home goal in four games led to Alan Pardew's men recording a first away defeat since he became Palace boss in January, the BBC reported.
For long periods in the game it looked like Southampton's woes in front of goal would continue, as Mane was guilty of several glaring misses in the first half. However, the introduction of substitute Ward-Prowse made a difference as his swelling run led to a first goal for Southampton in six hours and 27 minutes at St Mary's.
It was also Southampton's first victory in four league games. They remain in contention for a Champions League place despite their recent poor form.
Southampton boss Koeman said that they needed that three points because it wasn't the best game they played this season, insisting that they didn't create big chances, but adding that they tried and had the luck and the spirit to win the game.
Crystal Palace boss Pardew said that in the second half they were the better team and should have taken the lead, adding that they should have had a penalty as well, insisting that he believes Martin Atkinson should have given a penalty, Jose Fonte was going through the back of people all night.
Pardew claimed that it's so disappointing that his side had let in a bit of a soft goal, adding that Speroni is big enough to put up his hands up in the dressing room but insisted that they should have stopped it before they got anywhere near there.
